Determination of Relative Rice Leaf Water Content

实验原理:水稻叶片在非生物逆境胁迫条件下,叶片含水量有着较大的差异。
实验目的:通过叶片相对含水量的不同,检测水稻在非生物逆境下保持水分的能力,相对含水量越高,则水稻抗失水能力越强。

材料与试剂

  1. 冰盒
  2. 塑料袋
  3. 干净滤纸
  4. 50 ml离心管

仪器设备

  1. 干燥箱
  2. 分析天平
  3. 剪刀

实验步骤

  1. 在田间或温室用剪刀迅速剪取水稻完全展开叶片3-6片,至塑料袋中,再放置于冰盒中。
  2. 迅速带回实验室,将叶片取出摊开放置于干净滤纸上,按一定次序开始用分析天平称量鲜重记录为m1
  3. 将叶片放到50 ml离心管中,自来水浸泡2 h后将叶表面水分擦干,迅速称量记录m2
  4. 将叶片置于60 °C干燥箱10~15 h,至无水分丧失。称量干重记录m3
  5. 计算公式:
    叶片相对含水量(%) = (m1-m3)/(m2-m3)×100%
    叶片含水量(%) = (m1-m3)/m1×100%

注意事项

  1. 取样时试验样品和对照要相同时期,相同位置,大小相近。
  2. 从冰盒中取出后,将叶表面水分擦干后再称量。
  3. 若将叶片剪短称量,则片段大小应该大小相近,位置相同。
