实验原理及目的：冰冻切片适用于水分较多且容易收缩的材料。冰冻切片制片时间短，对样品组织损伤小，保护生物活性。但对切片技术、操作条件要求较高，被切细胞和形态结构不如石蜡切片好，因此具有一定的局限性。

材料与试剂

毛刷
载玻片
柑橘果皮
包埋剂 (O.C.T. Compound SAKURA Tissue-Tek, calalog number: 4583)
液氮
乙醇
乙酸
蔗糖
甘油

仪器设备

单面刀片
冰冻切片机 (Leica, model: CM1950)
注：CM1950冰冻切片机使用说明见附录。

实验步骤

材料处理
根据不同材料特性，可以选用不同的处理方式：
1.1
将材料分割成合适大小，不超过5 mm³，浸没于包埋剂1-5 min，时间不要太久，液氮速冻30 s-1 min或者放于冰冻切片机内速冻模块上速冻。
1.2
将材料分割成合适大小，不超过5 mm³，放于卡诺固定液 (纯乙醇15 ml，乙酸5 ml) 中，固定24 h后，由低浓度到高浓度换不同浓度保护剂 (常用蔗糖保护剂或甘油) 处理材料，再将材料浸没包埋剂中，液氮速冻30 s-1 min包埋。
注 (常用保护剂)：
10%蔗糖保护剂：10 g蔗糖 + 100 ml PBS
15%蔗糖保护剂：15 g蔗糖 + 100 ml PBS
修块、切片、粘片：将包埋好的材料粘在样品托上，用遇冷的刀片修整包埋块到合适大小进行切片，切片厚度一般10-14 µm。将切片粘附于干净的载玻片上。
注：
1)
OCT冻结后变得不透明，修块时要注意样品位置。
2)
可以用室温下的载玻片靠近切片，切片会自动粘附上。
染色 (选做)
用蒸馏水洗去切片上的包埋剂。将切片转入染色液中染色，用吸水纸擦拭干载玻片底面以及周围，置显微镜上观察。
