实验原理:转基因技术作为20世纪后期兴起的的生物工程技术,因其可以导入一个或多个外源基因从而改变一个至多个性状,并且可以克服柑橘有性杂交不亲和、珠心胚等影响。本实验的目的是在柑橘已有的遗传转化方法基础上,进行相应调整,建立高效转化方法,提高获得阳性芽的效率。本实验对转绿10天的早花柠檬上胚轴进行侵染,随后在25 °C、光照条件下以卡那抗生素作为筛选剂进行筛选培养,50 d后获得稳定抗性再生芽,对抗性再生芽进一步诱导生根,炼苗移栽至温室,整个实验周期约为4个月。
