Cytoplasmic Injection of mRNA In Monkey Embryos

摘要:利用显微操作仪将mRNA直接注射到受精卵的细胞质中,称为mRNA胞浆注射。相比于原核注射,胞浆注射的操作更加简便。结合低成本和易于设计的CRISPR/Cas技术,mRNA胞浆注射可以实现高效的基因编辑。在猴受精卵中注射sgRNA/Cas9 mRNA混合液,可以得到基因编辑的非人灵长类动物模型,为人类基因疾病的研究起到了重要的作用。

材料与试剂

  1. 3.5 cm 培养皿(CORNING,catalog number: 430165)
  2. 3.5 cm 操作皿(FALCON,catalog number: 353001)
  3. Borosilicate Glass(B100-75-10)

  5. 矿物油(Sigma,catalog number:M8410)

仪器设备

  1. 培养箱(Thermo Scientific, model:Forma 3111、Forma 371)
  2. 拉针仪(Sutter Instrument, model:P97)
  3. 断针仪(Narishige, model:MF-900)
  4. 倒置显微镜(Olympus, model:IX-73)
  5. 体视显微镜(Olympus, model:SZX7)
  6. IM300 Microinjector(Narishige)
  7. 一对显微操作臂(Narishige)
  8. 微量注射器(Narishige IM-9B)

实验步骤

实验流程

  1. 超排母猴及获取卵母细胞;
  2. 单精注射获取受精卵;
  3. 进行胚胎胞质内RNA注射,完成注射后,将胚胎移入培养碟中放置培养箱培养;
  4. 根据培养发育情况,将胚胎移植入受体体内;
  5. 做好详细记录。

mRNA注射

  1. 将TH3显微注射皿做好预热:取200 μl TH3液,在微操作碟内制备10-13 TH3 液滴,每滴体积约15-20 μl,并覆盖矿物油。
  2. 将HECM9aa 培养皿做好预热:取 200 μl HECM9aa培养液,在培养皿内制备10-13个液滴,每滴体积约15-20 μl,并覆盖矿物油。
  3. 用拉针仪将1 mm外径的薄壁硼硅酸毛细玻璃管(B100-75-10)拉制成需要的内外径后,在断针仪上断成外径为100-110 μm和内径为15-20 μm,打弯成20-30度的持卵针;
  4. 用拉针仪将1 mm外径的薄壁硼硅酸毛细玻璃管(BF100-78-10)拉制成需要的注射针,在断针仪上打弯20-30度;
  5. 将制作好的注射针倒扣在待注射的sgRNA中,以虹吸法吸入;
  6. 将制备的显微注射皿放置于载物台上,从低倍视野开始观察整个显微操作滴;
  7. 将持卵针安装于操作仪左侧的IM9B上,缓慢深入注射滴中,并调整与操作皿底部水平;
  8. 将注射针安装于操作仪右侧的IM300上,调整注射压力为30 psi、平衡压力0.1-0.3psi,缓慢深入注射滴中,并调整与操作皿底部水平,保持与持卵针在同一水平线;
  9. 单精注射完成后胚胎培养六个小时以上,在显微镜下检查原核形成情况,出现两个原核的胚胎视为正常受精;
  10. 用胚胎转移管将一组受精卵转移到操作皿中,操作尽可能在20-30 min内完成;
  11. 高倍镜下观察合子状态,确信观察到双原核而且形态良好;
  12. 用左侧持卵针抓取并固定合子后,推动右侧注射针透过透明带,进入卵胞质中,进行注射。观察注射针尖端是否通畅,若不通畅,将注射针在持卵针上轻轻碰击以增大注射针尖的直径,调整合适的注射压力;
  13. 完成注射后,将注射针从合子中迅速撤出,持卵针放开合子;
  14. 所有合子注射完毕后,应当立即移回HECM9aa培养液,置于37 °C,5% CO2 培养箱培养,等待胚胎移植。
