蛋白质降解是维持细胞蛋白质内稳态的重要机制。当无功能蛋白的溶酶体和1或蛋白酶体清除途径失效时，会形成聚合物，而这些聚合物可导致机体内潜发各种疾病，包括帕金森病和阿尔茨海默病。另一方面，通过影响细胞降解机制进而使靶蛋白发生明显降解，可作为一种治疗癌症、自身免疫和神经疾病等疾病新型治疗方法，拥有巨大的应用前景。相较于传统的小分子疗法，蛋白降解药物具有多种优势，例如其可扩大“可靶向"蛋白质组的使用范围，延长药代动力学持续时间;同时蛋白降解药物以催化的模式起作用，因此使用其进行治疗时，给予较低给药剂量即可达到较好效果。分子胶和蛋白降解靶向嵌合体( PROTAC)是该类新型小分子降解药物(如LYTAC. PHOTAC. PROTAC. 分子胶、AUTAC.疏水标记物)中最具有代表性的药物。
