材料与试剂

  1. 移液枪头

  11. 甘油三酯测定试剂盒 (GPO-PAP法) (中生北控)
  12. Tween-20 (阿拉丁, catalog number: T104863-100ml)
  13. BSA (阿拉丁, catalog number: A104912-100g)
  14. Brandford Reagent (Sigma, catalog number: B6916-500ML)
  15. 1× PBS缓冲液 (见溶液配方)

仪器设备

  1. 移液枪
  2. 酶标仪
  3. 研磨器 (Koktes)
  4. -80 °C冰箱 (Haier)
  5. 离心机 (Eppendorf, model: 5415R)
  6. Block Heater (Qilin Beier GL-150B)
  7. 高压蒸汽灭菌锅

实验步骤

  1. 样品准备:无论幼虫还是成虫,均以雌雄分开的方式收集样品 (组织样品需解剖,可以不分雌雄)。每组3个重复,每个重复5~10只。放入1.5 ml EP管后立即放入液氮,速冻后从液氮中取出管子,加入200 μl PBST (PBS + 0.05% Tween-20),将样品用研磨器研磨,可保存在-80 °C。
  2. 正式开始测定实验之前,预冷4 °C离心机,将干粉溶于10 ml反应溶液。将-80 °C冻存样品取出,瞬离样品溶液 (时间不宜长,否则使样品溶液不均匀,影响测定结果),然后置于冰上。
  3. 将上述溶液取出100 μl上清到另一1.5 ml EP管 (待测TAG,剩下100 μl用于测定蛋白) 置于冰上。
  4. 用于测定TAG的100 μl样品溶液置于70 °C反应10 min。热浴后的样品可重新置于-80 °C保存。
  5. 将热浴后的样品溶液取出10 μl与200 μl反应溶液。在标准曲线上取4个点,分别是0、0.0125、0.25、2.0 (mg),各取10 μl于反应溶液。将样品与标准曲线的混合液同时置于37 °C, 30 min后取出放室温。
  6. 将反应后的溶液室温12,000 × g, 2 min离心。移取离心后的反应溶液各190 μl于96孔酶标板中,记录样品位置待酶标仪测定 (Read Plate 540 nm)。
  7. 用于测蛋白的100 μl样品溶液4 °C,12,000 × g离心2 min。取10 μl上清于200 μl Brandford Reagent反应溶液中。配制蛋白标准样:0、0.0125、0.25、0.5、1、2 mg/ml BSA溶液,各取10 μl标准曲线溶液于200 μl Brandford Reagent反应溶液中,37 °C反应10 min,后取出放室温。取180 μl置于96孔板中,酶标仪测定蛋白浓度 (Read Plate 595 nm)。

溶液配方

  1. 1× PBS缓冲液
    磷酸缓冲盐缓冲液 (PBS)
    137 mmol/L NaCl
    2.7 mmol/L KCl
    10 mmol/L Na2HPO4
    2 mmol/L KH2PO4
    用800 ml蒸馏水溶解8 g NaCl,0.2 g KCl, 1.44 g Na2PHO4和0.24 g KH2PO4。用HCl调节溶液的pH值至7.4,加水至1 L。分装后在15 psi (1.05 kg/cm2) 高压蒸汽灭菌20 min,或通过过滤除菌,保存至室温
