Determination of Water Loss Rate of Excised-rice Leaves

实验原理:水稻叶片在离体状态下,通过气孔迅速散失水分。通过持续测定离体叶片鲜重,监测失水的速度。
实验目的:通过对离体叶片散失水分的情况,判断水稻叶片的保水能力,判断干旱相关表型是否与气孔调控相关。通常失水速率越快,水稻对干旱越敏感。

材料与试剂

  1. 塑料袋
  2. 冰盒
  3. 干净滤纸

仪器设备

  1. 分析天平
  2. 剪刀

实验步骤

  1. 在田间或温室用剪刀迅速剪取水稻完全展开叶片3-6片,至塑料袋中,再放置于冰盒中。
  2. 迅速带回实验室,将叶片取出摊开放置于干净滤纸上,每一样品设3个重复,按一定次序开始用分析天平称量并计数。
  3. 第一次称量记为0时刻,以后每半小时测一次鲜重,直至鲜重不再发生明显改变(约需6-8小时,失水率为60-70%)。
  4. 0时刻鲜重减去各时间点鲜重即为各时间点失水量,各时间点失水量再除以0时刻鲜重即为各时间点失水速率。
  5. 以时间点为横坐标,失水速率为纵坐标绘失水速率图,对每一时间点失水速率进行统计分析,判断两样品间失水速率是否有差异。

注意事项

  1. 取样时试验样品和对照要相同时期,相同位置,大小相近。
  2. 从冰盒中取出后,将叶表面水分擦干后再称量。
  3. 若将叶片剪短称量,则片段大小应该大小相近,位置相同。
