Structural and Functional role of B2-GPI in Anti-phospholipid Syndrome:

β2-glycoprotein I (β2GPI) is an abundant multidomain plasma protein that plays various roles in the clotting and complement cascades. Beta-2-Glycoprotein I (β2GPI) is a unique five domain protein comprising four similar complement control protein (CCP)-like domains (DI-DIV) and one different domain (DV) with a large lysine. It is a soluble blood protein with a circulating concentration of 0.2 mg/ml and a molecular weight of 50kDa with glycosylation. β2GPI has many proposed functions and roles within the body including the regulation of complement and hemostasis. It is also the main target of anti- phospholipid antibodies (aPL) in the acquired coagulopathy known as antiphospholipid syndrome (APS). Being an important target against APS, the structure and function of B2-GPI should be explored in detail to make it as therapeutic target to treat APS disease.
We are using the state-of-the-art biochemical and biophysical methods (Biochemical methods, SPR, X-ray crystallography, SAXS, Cryo-EM, smFRET) in our lab to study the dynamic behaviors, structure, and functions of proteins involved in antiphospholipid syndrome.
