Equipment

pH试纸；600ml、1500ml烧杯或烧瓶；量筒；搅拌器。

Procedure

配制EDTA储存液（以500ml，0.5M EDTA为例）

称取93.05gEDTA二钠盐
溶解在400ml去离子水中，用1M NaOH溶液调节pH至8.0使EDTA完全溶解
定容至500ml，保持溶液pH为8.0

配制50XTAE储存液

称量242gTris base
溶解在400ml去离子水中
加入57.1ml冰醋酸，100ml，0.5M EDTA（pH 8.0）
定容至1L
常温储存，(此时pH约为8.5)

配制TAE工作液（40mM Tris醋酸 1mM EDTA）

稀释50*TAE储存液至1*TAE工作液
